There are several ways of getting your Dwellers’ happiness up in Fallout Shelter, as well as keeping them content while they’re working in your Vault. Here’s everything you can do:

Keep your resource productions above the limit. Not having enough water and food will impact their health as well, and having the lights out of their assigned workplaces brings Dweller down.
Build a Radio Studio and transmit inside the Vault instead of looking for Dwellers in the Wasteland. If you also place high Charisma dwellers inside the studio, you’ll greatly boost Dwellers’ happiness.
Assign Dwellers to a correct job according to their highest SPECIAL stat.
Assign them to Living Quarters and get them to talk to potential partners.
If you manage to get a successful rush, you’ll raise the overall happiness of that room by 10%.
If your Dwellers are hurt or affected by radiation, use RadAways or Stimpaks to heal them and raise their happiness.
